Its believed that the stronger the pre-out voltage, the better the signal quality to your amp. Most amp pre-out input voltages can recieve from approx. .5v to 5v. This is where the gain knobs comes into effect. A scenario: "your deck oututs signal at 4volts, your amp recieves it at 4volts. your gain adjust goes from .5 to 5volts. Its basically a sensitivity knob for recieving the signal. If the signal is weaker(closer to .5) you want to turn the gain as close to .5v to get a strong signal. Where the gain really comes into effect is when you have a 2v - 4v signal and you turn your gain towards .5v, you amplify the incomming signal so your system will play louder. Distortion can occur and speaker damage so be careful when reaching max output volume from your headunit. The benefits of having a deck with 4v pre-outs over 2.5 is you do not need to turn your gain up as high to achieve the same level of sound, the quality of signal is a little better and less signal loss is achieved with 4v pre outs when accompanied with quality signal cables.
